Overview

NT219 + cetuximab is a combination therapy under investigation for the treatment of recurrent or metastatic squamous cell carcinoma of the head and neck (SCCHN). NT219 is a first-in-class, novel small molecule developed to overcome tumor resistance by targeting and degrading insulin receptor substrate 1 (IRS1) and insulin receptor substrate 2 (IRS2) and inhibiting signal transducer and activator of transcription 3 (STAT3), both key oncogenic drivers involved in drug resistance pathways. By blocking these pathways, NT219 aims to resensitize tumors to standard-of-care therapies such as EGFR inhibitors like cetuximab. Cetuximab is a monoclonal antibody that targets the epidermal growth factor receptor (EGFR). The combination is being evaluated in Phase 2 clinical trials for its efficacy in overcoming resistance to EGFR blockade in SCCHN[1][4][5][6].
