Overview

NTX-1088 + pembrolizumab is an investigational combination therapy for advanced solid tumors. NTX-1088 is a first-in-class monoclonal antibody targeting the poliovirus receptor PVR (CD155), a protein associated with resistance to PD-1/PD-L1 checkpoint blockade. By blocking PVR, NTX-1088 restores DNAM1 (CD226)-mediated activation of T and NK cells and inhibits suppressive signaling through TIGIT and CD96. Pembrolizumab, marketed as Keytruda, is a monoclonal antibody that blocks PD-1, thereby enhancing T cell–mediated antitumor immunity. The combination is designed to synergistically enhance antitumor immune responses, especially in tumors expressing PVR and resistant to PD-1 inhibition. Currently, clinical investigation is in Phase 1 for advanced solid malignancies[1][2][3][4][5][7].
