Overview

NUV-868 + enzalutamide is an oral combination therapy under investigation for the treatment of advanced solid tumors, particularly metastatic castration-resistant prostate cancer (mCRPC). NUV-868 is a BD2-selective BET (bromodomain and extra-terminal domain) inhibitor, which targets epigenetic regulators such as BRD4, thereby modulating genes that control tumor growth and differentiation. Enzalutamide is an androgen receptor inhibitor that blocks the androgen receptor signaling pathway, which is critical for the progression of prostate cancer. The combination is being studied to address tumor resistance mechanisms and potentially deliver synergistic antitumor activity by targeting both epigenetic and androgen receptor-driven growth pathways[1][3][5][7].
