Overview

**NUV-868 + olaparib** is a combination investigational therapy for advanced solid tumors. NUV-868 is a selective inhibitor of BET (bromodomain and extra-terminal domain) proteins, specifically targeting the BD2 domain of BRD4, a key protein involved in gene transcription and tumor cell growth. Olaparib is a clinically approved inhibitor of poly(ADP-ribose) polymerase (PARP), a critical DNA repair enzyme. Together, the combination aims to enhance anti-tumor efficacy by concurrently interfering with tumor growth transcriptional programs and inhibiting DNA repair, potentially improving outcomes in patients with resistant or advanced cancers such as ovarian, pancreatic, metastatic castration-resistant prostate cancer (mCRPC), and triple-negative breast cancer.
