Drug intelligence / Profile preview
NV103 is a preclinical-stage targeted nanoparticle drug conjugate (nADC) developed by NanoValent Pharmaceuticals in collaboration with the University of Southern California and Children's Hospital Los Angeles. It consists of the topoisomerase I inhibitor irinotecan encapsulated within hybrid polymerized liposomal nanoparticles (HPLNs) that are covalently conjugated with a human monoclonal antibody targeting CD99 (p30/32MIC2). CD99 is a surface glycoprotein highly expressed on Ewing sarcoma cells and certain brain tumors like glioblastoma multiforme. By utilizing active antibody-mediated targeting, NV103 aims to deliver high concentrations of chemotherapy directly to tumor cells while minimizing systemic toxicity and bypassing the blood-brain barrier. Preclinical studies have demonstrated superior efficacy compared to both free irinotecan and untargeted liposomal formulations in models of Ewing sarcoma and glioblastoma.
