NVR 3-778 + peginterferon alfa-2a is a combination therapy composed of NVR 3-778, a small-molecule hepatitis B virus (HBV) capsid assembly modulator, and peginterferon alfa-2a, a pegylated recombinant human interferon used as an immunomodulatory and antiviral agent. NVR 3-778 inhibits HBV replication by targeting and modulating the core protein, interfering with viral capsid assembly and blocking pregenomic RNA encapsidation and subsequent production of infectious HBV DNA and RNA particles. Peginterferon alfa-2a activates human type I interferon receptors, leading to JAK/STAT pathway signaling and induction of innate antiviral gene expression. The combination has been explored in clinical contexts for the treatment of chronic hepatitis B, aiming to provide additive or synergistic antiviral effects by targeting different stages of the HBV life cycle[1][2][3][5].
