NY-ESO-1 + alum polysaccharide + HH2 is an investigational combination cancer vaccine composed of a recombinant NY-ESO-1 antigen, an alum-based polysaccharide adjuvant, and HH2 (a peptide-based immune adjuvant). NY-ESO-1 is a cancer-testis antigen highly immunogenic and widely expressed in multiple solid tumors but rarely in normal adult tissues, making it an attractive cancer immunotherapy target. The vaccine is designed to elicit both cellular (CD4+ and CD8+ T cell) and humoral immune responses against NY-ESO-1-expressing tumor cells. The alum polysaccharide component acts as an adjuvant to enhance antigen presentation and immune activation, while HH2 further stimulates the innate immune system, likely through TLR pathways (such as via TLR2 and TLR4) and chemotactic mechanisms to potentiate vaccine-induced immunity. The combination aims to induce a tumor-specific immune response capable of targeting and killing NY-ESO-1-expressing cancer cells, mainly in solid tumors such as melanoma, lung cancer, ovarian cancer, and others. This vaccine formulation is currently in clinical or preclinical development stages and is part of cancer immunotherapy research, with no current approvals for clinical use[1][2][4].
