Overview

NY-ESO-1 + CpG 7909 is a combination immunotherapeutic approach used primarily as a cancer vaccine. It combines the tumor antigen NY-ESO-1, a highly immunogenic cancer/testis antigen expressed in various tumors but not in most normal tissues, with CpG 7909 (also known as PF3512676), a synthetic oligodeoxynucleotide that acts as an immune adjuvant by stimulating Toll-like receptor 9 (TLR9). The combination is designed to enhance anti-tumor immune responses by promoting robust activation and expansion of NY‑ESO‑1-specific cytotoxic T lymphocytes. Clinical studies have shown that this combination can induce strong cellular and humoral immune responses against tumors expressing NY‑ESO‑1, particularly in melanoma and other solid cancers[1][3][5]. The vaccine is typically administered with Montanide ISA 720 as an additional adjuvant to further boost immunogenicity[1]. This strategy aims to harness the body’s own immune system for targeted destruction of cancer cells.
