Overview

NY-ESO-1 ISCOMATRIX vaccine is a cancer immunotherapy developed by the Ludwig Institute for Cancer Research in collaboration with CSL Limited. The vaccine comprises a full-length recombinant NY-ESO-1 protein, a cancer-testis antigen frequently expressed in advanced melanomas but absent in most normal tissues. It is formulated with ISCOMATRIX, a proprietary saponin-based adjuvant designed to enhance the magnitude and breadth of the immune response. Administered intramuscularly, the vaccine aims to stimulate both antibody production and T-cell mediated immunity (specifically CD4+ and CD8+ T cells) against tumor cells. Clinical evaluation has primarily focused on patients with resected or advanced malignant melanoma, where it has demonstrated significant immunogenicity and a favorable safety profile.
