Overview

NY-ESO-1 OLP4 + Montanide is a therapeutic cancer vaccine composed of four long overlapping synthetic peptides (OLP4) derived from the immunogenic cancer-testis antigen NY‑ESO‑1, combined with the adjuvant Montanide ISA 51. The vaccine is designed to stimulate both humoral and cellular immune responses against tumors expressing NY‑ESO‑1. The overlapping peptide approach covers most T cell epitopes within residues 79–173 of the protein, maximizing immunogenicity and minimizing HLA restriction[2][3][4]. Montanide acts as an incomplete Freund’s adjuvant to enhance antigen presentation and immune activation. This combination has been studied primarily in ovarian epithelial carcinoma, fallopian tube carcinoma, and primary peritoneal carcinoma[3][5]. Clinical trials have shown that adding Montanide increases CD8+ T-cell cross priming and enhances T-cell avidity for the target antigen[1].
