NY-ESO-1 OLP4 vaccine + poly-ICLC + montanide is a therapeutic cancer vaccine combination designed to induce immune responses against tumors expressing the cancer-testis antigen NY‑ESO‑1. The formulation consists of four overlapping long peptides (OLP4) derived from the immunogenic region of the NY‑ESO‑1 protein, combined with two adjuvants: Montanide ISA 51 VG (a water-in-oil emulsion that enhances antigen presentation) and polyinosinic-polycytidylic acid stabilized with poly-L‑lysine and carboxymethylcellulose (poly‑ICLC), a synthetic double-stranded RNA that acts as a toll-like receptor 3 agonist to further stimulate innate immunity. This combination aims to activate both CD8+ and CD4+ T cell responses against tumor cells expressing NY‑ESO‑1, particularly in patients with epithelial ovarian, fallopian tube, or primary peritoneal carcinoma in remission after standard therapy[5][6]. The approach is intended to overcome HLA restriction by covering most immunogenic epitopes within residues 79–173 of the protein[6].
