The NY-ESO-1 peptide vaccine is a cancer immunotherapy developed by Roswell Park Comprehensive Cancer Center. It targets the NY-ESO-1 (New York Esophageal Squamous Cell Carcinoma 1) cancer-testis antigen, which is frequently expressed in various solid tumors, including ovarian, fallopian tube, and primary peritoneal cancers. The vaccine consists of the NY-ESO-1-derived long peptide ESO157–170 (sequence: SLLMWITQCFLPVF), which possesses dual MHC Class I and Class II specificities. It is formulated with the adjuvant Montanide ISA51 (incomplete Freund's adjuvant) to enhance the immune response. The vaccine aims to stimulate HLA-A2- and HLA-A24-restricted CD8+ cytotoxic T cell responses, HLA-DP4-restricted CD4+ T helper cell responses, and humoral antibody responses against tumor cells expressing the NY-ESO-1 antigen. It is currently being evaluated in Phase I clinical trials, often in combination with chemotherapy or epigenetic agents like decitabine.
