Overview

NY-ESO-1 protein + imiquimod is an investigational cancer vaccine developed by the Ludwig Institute for Cancer Research for the treatment of malignant melanoma. The vaccine consists of a recombinant full-length NY-ESO-1 protein, a cancer-testis antigen expressed in approximately 20-30% of melanomas, administered in combination with imiquimod, a Toll-like receptor 7 (TLR7) agonist that serves as an adjuvant. The treatment regimen involves the topical application of imiquimod cream (Aldara) to precondition the skin, followed by the intradermal injection of the NY-ESO-1 protein. This combination is designed to elicit both humoral and cellular immune responses against NY-ESO-1-expressing tumor cells. It has been evaluated in Phase 1 clinical trials for patients with resected Stage IIB, IIC, and III malignant melanoma.
