Overview

NY-ESO-1 protein + poly-ICLC + montanide is a therapeutic cancer vaccine combination designed to induce robust humoral and cellular immune responses against tumors expressing the NY‑ESO‑1 antigen. The vaccine consists of the full-length recombinant NY‑ESO‑1 protein (a highly immunogenic cancer/testis antigen), combined with two adjuvants: * Polyinosinic-polycytidylic acid stabilized with poly-L‑lysine and carboxymethylcellulose (poly‑ICLC), a synthetic double-stranded RNA that acts as an agonist for Toll-like receptor 3 (TLR3) and melanoma differentiation-associated gene 5 (MDA5), stimulating innate immunity. * Montanide ISA 51 VG, an oil-based emulsion used to enhance antigen presentation. This combination has been evaluated in phase I/II clinical trials primarily in patients with high-risk resected melanoma. The regimen is generally well tolerated, induces strong antibody and CD4+ T-cell responses specific for NY‑ESO‑1, and—when combined with montanide—enhances CD8+ T-cell cross priming and avidity. The approach aims to prevent recurrence by activating both arms of adaptive immunity against residual tumor cells expressing the target antigen[1][2][5].
