Overview

NY-ESO-1 T-Cell Receptor (TCR) therapy is a cell therapy that involves engineering T cells to express a high-affinity T-cell receptor (TCR) specific for the NY-ESO-1 cancer-testis antigen. These engineered T cells are designed to recognize and destroy cancer cells presenting the NY-ESO-1 peptide in the context of HLA-A2. The therapy has shown feasibility and antitumor responses in patients with multiple myeloma in a Phase I/II clinical trial. A simplified "single editing" (SE) approach, which disrupts only the endogenous TCR alpha chain, has been developed to improve safety and manufacturing feasibility, reducing alloreactivity while maintaining potent antitumor activity. The therapy was developed by Adaptimmune and Penn Medicine.
