Overview

This investigational combination immunotherapy, developed by the Jonsson Comprehensive Cancer Center at UCLA, consists of autologous peripheral blood mononuclear cells (PBMCs) genetically engineered with a retroviral vector to express a T-cell receptor (TCR) specific for the NY-ESO-1 cancer-testis antigen (specifically the 157-165 epitope). The treatment protocol involves the adoptive transfer of these engineered T-cells following a non-myeloablative lymphodepleting conditioning regimen of cyclophosphamide and fludarabine. To enhance the expansion, persistence, and anti-tumor activity of the transferred cells, the regimen includes the administration of a vaccine composed of autologous dendritic cells pulsed with the NY-ESO-1 peptide, along with systemic aldesleukin (IL-2). This multi-component approach is designed to treat advanced NY-ESO-1-positive malignancies, such as melanoma and various sarcomas, by leveraging both passive and active immunization strategies.
