Overview

**NZV930 + PDR001 + NIR178** is an experimental combination regimen of three investigational immunotherapy agents developed for advanced malignancies. NZV930 is a monoclonal antibody targeting CD73 (ecto-5'-nucleotidase) to reduce adenosine-mediated immunosuppression in the tumor microenvironment. PDR001, also known as spartalizumab, is a monoclonal antibody that inhibits the programmed cell death 1 (PD-1) receptor, thereby enhancing T-cell mediated anti-tumor immunity. NIR178, also called taminadenant, is a small molecule antagonist of the adenosine A2A receptor (ADORA2A), aiming to further reverse immunosuppression driven by extracellular adenosine. The combination was studied in adult patients with advanced solid tumors, including but not limited to non-small cell lung cancer, triple-negative breast cancer, pancreatic ductal adenocarcinoma, colorectal cancer (microsatellite stable), ovarian cancer, renal cell carcinoma, and metastatic castration-resistant prostate cancer. The Phase I/Ib trial evaluated safety, tolerability, pharmacokinetics, and preliminary anti-tumor activity. The recommended Phase II dose was NZV930 600 mg IV every 2 weeks, PDR001 (spartalizumab) 400 mg IV every 4 weeks, and NIR178 (taminadenant) 240 mg orally twice a day. The trial was terminated early due to insufficient efficacy in the expansion arms and not for safety reasons[1][3][5][7].
