**OA@CuMnCs** is a preclinical, copper-manganese biomineralized oncolytic adenovirus developed for colorectal cancer immunotherapy. The construct coats an oncolytic adenovirus with copper and manganese ions to reduce immune clearance and improve tumor delivery. Copper-associated activity reduces tumor-cell programmed death ligand 1 expression and supports antigen presentation, dendritic-cell maturation, immunogenic cell death, and conversion of immunologically cold tumors to hot tumors. Manganese reacts with endogenous hydrogen peroxide to generate oxygen, alleviating hypoxia and supporting adenoviral replication and tumor-cell lysis.
