oAd + LP-GE-PS is an experimental gene therapy and oncolytic virus formulation designed for the systemic treatment of EGFR-positive cancers. It consists of an oncolytic adenovirus (oAd) engineered to coexpress short-hairpin RNA (shRNA) against Wnt5 (shWnt5) and the tumor-suppressive protein decorin. To facilitate systemic delivery, the virus is encapsulated in a dual-functionalized lipid envelope (LP). This envelope is modified with the GE11 peptide (GE) to target the epidermal growth factor receptor (EGFR) and a pH-responsive sulfamethazine-based polymer (PUSSM, denoted as PS) that triggers the release of the viral payload within the acidic tumor microenvironment. This multi-component system is designed to minimize hepatic sequestration and maximize intratumoral accumulation and penetration, thereby enhancing therapeutic efficacy while reducing systemic toxicity.
