**Oblimersen + dexamethasone** is a combination regimen consisting of *oblimersen sodium* (an antisense oligonucleotide) and *dexamethasone* (a synthetic glucocorticoid). Oblimersen specifically binds to Bcl-2 mRNA, leading to decreased production of the anti-apoptotic Bcl-2 protein. This mechanism aims to sensitize malignant plasma cells (especially in multiple myeloma) that often overexpress Bcl-2, resulting in resistance to apoptosis and chemotherapy. Dexamethasone serves as an antineoplastic and immunosuppressive agent, commonly used in the treatment of hematologic malignancies. Experimental and clinical data indicate that combining oblimersen with dexamethasone enhances apoptosis in resistant myeloma cells, though a phase III trial in relapsed/refractory multiple myeloma did not show a statistically significant benefit over dexamethasone alone[1][2][3][4].
