A combination regimen of the antisense oligonucleotide oblimersen and the antibody-drug conjugate gemtuzumab ozogamicin for CD33-positive acute myeloid leukemia. Oblimersen is a phosphorothioate antisense DNA oligonucleotide targeting BCL2 mRNA to reduce BCL-2 protein and enhance apoptosis. Gemtuzumab ozogamicin is a humanized anti-CD33 monoclonal antibody conjugated to the cytotoxic agent calicheamicin; upon binding CD33 on myeloid blasts, it is internalized and releases calicheamicin to induce DNA double-strand breaks and cell death. This combination was studied primarily in relapsed AML in older adults to potentiate GO efficacy by downregulating anti-apoptotic BCL-2.
