**OC125F(ab')2-DTPA-153Sm** is a preclinical CA125-targeted radioimmunotherapy agent for epithelial ovarian cancer. It consists of the murine anti-CA125 monoclonal antibody OC125 as an F(ab')2 antibody fragment, conjugated through diethylenetriaminepentaacetic acid (DTPA) to the beta-emitting therapeutic radionuclide samarium-153. Binding of the OC125 fragment to mucin-16 on ovarian cancer cells is intended to localize samarium-153 radiation to tumor tissue. In SHIN-3 ovarian adenocarcinoma xenograft-bearing nude mice, the DTPA conjugate showed limited serum stability, with approximately 50% of samarium-153 remaining antibody-bound, tumor uptake of 4.53% injected activity per gram at 24 hours, and unfavorable liver and bone distribution relative to a more stable CITCDTPA comparator.
