This combination therapy regimen consists of ociperlimab (an anti-TIGIT monoclonal antibody) and tislelizumab (an anti-PD-1 monoclonal antibody) administered in conjunction with concurrent chemoradiotherapy (cCRT). The cCRT component typically includes platinum-based chemotherapy (cisplatin or carboplatin) and etoposide, along with thoracic radiation therapy. Developed by BeiGene, the regimen was investigated for the treatment of patients with untreated limited-stage small cell lung cancer (LS-SCLC) in the Phase 2 trial NCT04952597. Ociperlimab targets the T-cell immunoreceptor with Ig and ITIM domains (TIGIT) to enhance immune response, while tislelizumab blocks the programmed cell death protein 1 (PD-1) pathway. In 2023, BeiGene discontinued the clinical development of ociperlimab in lung cancer following a futility analysis recommendation from an independent data monitoring committee.
