A combination therapy composed of **octacalcium phosphate** (OCP), **vascular endothelial growth factor A (VEGFA) plasmid DNA**, and **autologous bone (autobone)**, designed as a gene-activated bone substitute for bone tissue regeneration. OCP acts as a biocompatible scaffold that delivers naked plasmid DNA encoding VEGFA, thereby promoting localized angiogenesis and osteoinduction. The plasmid DNA is released from the scaffold and taken up by local cells, where VEGFA expression is upregulated, stimulating neovascularization and supporting new bone formation. Autobone provides viable osteogenic cells and additional structural support. This approach leverages both the physical properties of the scaffold and the biological effects of gene therapy to enhance bone healing, particularly in large or critically sized bone defects. Preclinical evidence suggests enhanced vascularization and bone tissue regeneration compared to controls. Clinical trials have been initiated for device registration, indicating translational development for orthopedic or reconstructive applications[1][4].
