Drug intelligence / Profile preview
OD-001 + vedolizumab is an investigational combination regimen being developed by Odyssey Therapeutics for the treatment of moderate-to-severe ulcerative colitis. The regimen combines OD-001 (also known as OD-07656), an oral small molecule scaffolding inhibitor of receptor-interacting serine/threonine-protein kinase 2 (RIPK2), with vedolizumab, an approved humanized IgG1 monoclonal antibody targeting the α4β7 integrin. OD-001 blocks upstream innate immune activation by preventing RIPK2 scaffolding with XIAP, thereby inhibiting NOD1/NOD2-mediated inflammatory signaling. Vedolizumab selectively inhibits adaptive immune cell trafficking to the gut by blocking the interaction of α4β7 integrin with mucosal addressin cell adhesion molecule-1 (MAdCAM-1). This combination strategy aims to target complementary innate and adaptive immune pathways to enhance the induction of response and remission in patients with ulcerative colitis.
