Overview

Ofranergene obadenovec (VB-111, also known as ofra-vec) is a non-replicating adenoviral vector-based gene therapy designed for cancer treatment. It carries a pro-apoptotic human Fas-chimera transgene under the control of a modified murine PPE-1-3x promoter and is specifically expressed in angiogenic endothelial cells. The drug exerts dual mechanisms—vascular disruption (anti-angiogenesis) and immune activation—by inducing apoptosis in tumor vasculature and stimulating CD8+ T-cell infiltration into tumors[1][4][5]. Paclitaxel is a standard chemotherapeutic agent that stabilizes microtubules, inhibiting cell division. The combination has been investigated primarily for recurrent platinum-resistant ovarian cancer in multiple phase III clinical trials[1][5][8]. Ofranergene obadenovec was developed by VBL Therapeutics.
