OGX-427 + docetaxel is a combination therapy consisting of OGX-427 (also known as apatorsen), a second-generation antisense oligonucleotide targeting heat shock protein 27 (HSP27), and docetaxel, a taxane-class cytotoxic chemotherapy. OGX-427 inhibits HSP27 expression, which is known to contribute to cancer cell survival, chemoresistance, and disease progression. By suppressing HSP27, OGX-427 promotes apoptosis and enhances the efficacy of chemotherapeutic agents including docetaxel. The combination has been studied in advanced solid tumors, most notably in metastatic castration-resistant prostate cancer, as well as in urothelial cancer and other settings. In published clinical trials, the combination showed evidence of antitumor activity, including partial responses and PSA declines, and was generally well tolerated with infusion reactions being the primary dose-limiting toxicity[3][4][8].
