Overview

OH2 + irinotecan is a combination therapy under investigation for solid tumors, particularly advanced gastrointestinal cancers. OH2 is an oncolytic virus based on a genetically modified herpes simplex virus type 2 (HSV-2) engineered to selectively replicate in tumors. It also delivers the gene encoding human granulocyte macrophage colony-stimulating factor (GM-CSF) to stimulate a stronger antitumor immune response. Irinotecan is a topoisomerase I inhibitor used mainly in the treatment of cancer, particularly colorectal and gastrointestinal cancers. The combination aims to enhance the direct cytolytic effect of OH2 with the cytotoxic and antiproliferative effects of irinotecan. Both agents may act synergistically, leveraging oncolytic viral therapy and chemotherapy to improve responses in difficult-to-treat cancers[4].
