olanzapine + amantadine

Olanzapine + amantadine is a combination of two pharmaceutical drugs, each with distinct mechanisms of action. Olanzapine is an atypical antipsychotic (second-generation) primarily used for the treatment of schizophrenia, bipolar disorder, and agitation associated with these conditions. It acts mainly as an antagonist at dopamine D2 and serotonin 5-HT2A receptors, among others. Amantadine is an adamantane derivative used as both an antiviral agent (against influenza A) and as a treatment for Parkinson's disease and drug-induced extrapyramidal symptoms; it increases dopamine release and inhibits its reuptake in the brain. The combination has been studied for mitigating olanzapine-induced weight gain in patients with psychiatric disorders; evidence suggests that amantadine may help reduce weight gain associated with olanzapine therapy[4]. However, this use is not routine clinical practice.
