Overview

This therapeutic regimen is a combination of the PARP inhibitor olaparib, the anti-EGFR monoclonal antibody cetuximab, and radiation therapy, specifically evaluated for the treatment of locally advanced squamous cell carcinoma of the head and neck (HNSCC) in patients with heavy smoking histories. Olaparib acts as a radiosensitizer by inhibiting PARP-1 and PARP-2 enzymes, which are critical for repairing single-strand DNA breaks, thereby enhancing the lethal effects of radiation-induced DNA damage. Cetuximab targets the epidermal growth factor receptor (EGFR), which is frequently overexpressed in HNSCC and associated with resistance to radiation. The combination aims to overcome treatment resistance and improve survival outcomes in a high-risk patient population where standard chemoradiation may be less effective.
