Overview

**Olaparib + dacarbazine** is a combination of two small molecule drugs: **olaparib**, a potent inhibitor of poly (ADP-ribose) polymerase (PARP), and **dacarbazine**, an alkylating agent that induces DNA damage. Olaparib impairs the cellular repair of DNA lesions, especially via inhibition of the base excision repair pathway, making tumor cells more susceptible to the effects of DNA-damaging agents like dacarbazine. The combination aimed to exploit this synergy for increased cytotoxicity in cancer cells, particularly in advanced solid tumors and melanoma. In a phase I clinical study, the combination at certain doses was tolerable but did not show clinical benefit over dacarbazine monotherapy for chemonaive melanoma patients[1][2]. The regimen has been tested mainly in the clinical trial context, and its use is investigational.
