Olaparib + durvalumab is a combination therapy of two pharmaceutical agents used primarily in oncology clinical trials. Olaparib is a small molecule inhibitor of poly [ADP-ribose] polymerase (PARP), specifically targeting PARP1 and PARP2, which are enzymes involved in DNA repair. By inhibiting these enzymes, olaparib exploits defects in DNA repair pathways—particularly in tumors with BRCA mutations—leading to cancer cell death[1][5][6]. Durvalumab is a monoclonal antibody that targets the programmed death-ligand 1 (PD-L1), blocking its interaction with PD-1 and CD80 on T cells, thereby enhancing anti-tumor immune responses[3][6]. The combination has been investigated for synergistic effects, as PARP inhibition may increase tumor immunogenicity and sensitize tumors to immune checkpoint blockade. This regimen has been studied in various cancers including breast cancer, ovarian cancer, endometrial cancer, small cell lung cancer (SCLC), and non-small-cell lung carcinoma (NSCLC)[2][3][6][7][8].
