Overview

Olaparib + durvalumab + tremelimumab is a combination therapy under investigation for the treatment of solid tumors with mutations in homologous recombination repair genes and other cancers such as ovarian, fallopian tube, or primary peritoneal cancer. Olaparib is a small molecule PARP inhibitor that impairs DNA repair in tumor cells with deficient homologous recombination. Durvalumab is a human monoclonal antibody targeting programmed cell death ligand 1 (PD-L1), blocking its interaction with PD-1 and CD80 to enhance T-cell mediated antitumor responses. Tremelimumab is a monoclonal antibody directed against cytotoxic T-lymphocyte-associated protein 4 (CTLA-4), promoting immune activation by inhibiting an immune checkpoint. The combination aims to exploit both direct tumoricidal effects and enhanced antitumor immunity[1][2][3].
