Overview

This is a combination therapy consisting of three agents: olaparib, a PARP inhibitor; durvalumab, an anti-PD-L1 monoclonal antibody; and UV1, a therapeutic cancer vaccine targeting telomerase. Olaparib inhibits poly(ADP-ribose) polymerase enzymes involved in DNA repair, leading to increased tumor neoantigen formation and modulation of the tumor microenvironment. Durvalumab blocks PD-L1 on tumor cells, enhancing T-cell mediated immune responses against cancer. UV1 is designed to induce an immune response against telomerase-expressing tumor cells by stimulating T-cell activity. The combination aims to leverage DNA damage-induced immunogenicity with checkpoint inhibition and active vaccination for synergistic antitumor effects. This regimen is under investigation as maintenance therapy following platinum-based chemotherapy in patients with recurrent ovarian cancer without BRCA mutations[1][2][4][8].
