Olaptesed pegol + bevacizumab is a combination therapy under investigation for glioblastoma. Olaptesed pegol (NOX-A12) is a PEGylated antisense oligonucleotide that acts as a selective inhibitor of CXCL12 (stromal cell-derived factor 1), disrupting tumor-supporting microenvironments and impeding cancer cell migration, proliferation, and immune evasion. Bevacizumab is a monoclonal antibody targeting vascular endothelial growth factor A (VEGF-A), thus inhibiting tumor angiogenesis. As a combination with radiotherapy, this regimen showed significantly improved median overall survival and response rates compared to standard of care in newly diagnosed, chemotherapy-resistant glioblastoma with measurable residual tumor after surgery.
