olaratumab + doxorubicin

**Olaratumab + doxorubicin** is a combination therapy consisting of the monoclonal antibody olaratumab and the cytotoxic anthracycline doxorubicin. Olaratumab is a human IgG1 monoclonal antibody that selectively binds and blocks platelet-derived growth factor receptor alpha (PDGFRα), thereby inhibiting PDGFRα-mediated signaling involved in tumor growth, proliferation, and angiogenesis. Doxorubicin is a DNA intercalating agent and a topoisomerase II inhibitor that induces DNA breaks, leading to apoptosis in dividing cells. This combination was developed and approved for the treatment of advanced soft tissue sarcoma (STS) in adults not amenable to curative surgery or radiotherapy and not previously treated with doxorubicin. Initial clinical studies suggested a significant improvement in overall survival and progression-free survival with the combination, but the confirmatory phase 3 trial failed to replicate these findings, leading to regulatory withdrawal of olaratumab in many markets.
