Overview

Olinvacimab + pembrolizumab is a combination therapy investigated primarily for metastatic triple-negative breast cancer (mTNBC) and recurrent glioblastoma multiforme (rGBM)[1][2][5][7]. Olinvacimab is a fully humanized monoclonal antibody that binds to Vascular Endothelial Growth Factor Receptor 2 (VEGFR2), inhibiting angiogenesis and tumor growth by blocking VEGF ligands—including VEGF-A, VEGF-C, and VEGF-D—from activating VEGFR2[1][2][5]. Pembrolizumab is a monoclonal antibody that targets Programmed cell death protein 1 (PD-1) on T cells, antagonizing its interaction with PD-L1 and PD-L2, which releases the inhibition of immune response and enhances T-cell mediated anti-tumor activity[1][4][5][6][10]. The combination is tested for synergistic effects between anti-angiogenic and immune checkpoint blockade.
