Overview

Olutasidenib + cytarabine is an investigational combination treatment composed of olutasidenib, a potent and selective small molecule inhibitor of mutant isocitrate dehydrogenase 1 (IDH1), combined with cytarabine, a nucleoside analog antimetabolite chemotherapy. Olutasidenib targets and inhibits the neomorphic enzymatic activity of mutant IDH1, thereby reducing the production of the oncometabolite 2-hydroxyglutarate, which contributes to leukemogenesis. Cytarabine incorporates into DNA during replication, inhibiting DNA synthesis and inducing apoptosis in rapidly proliferating cells. The combination has been studied in patients with relapsed or refractory acute myeloid leukemia (AML) harboring an IDH1 mutation, including within a multi-arm phase 1/2 clinical trial. The purpose of the combination is to assess synergistic or additive efficacy in high-risk or refractory AML populations. Both agents are administered via established dosing regimens within clinical trials, but the combination is not yet approved for standard care.
