Drug intelligence / Profile preview
Olvi-Vec (olvimulogene nanivacirepvec) in combination with platinum-based chemotherapy and an immune checkpoint inhibitor (ICI) is an investigational immunotherapeutic regimen being evaluated for the treatment of advanced or metastatic solid tumors, particularly recurrent non-small cell lung cancer (NSCLC). Olvi-Vec is a genetically modified, oncolytic vaccinia virus (Lister strain) designed to selectively infect, replicate within, and lyse cancer cells. This oncolysis releases tumor-associated antigens and pathogen-associated molecular patterns, converting the tumor microenvironment from immunologically "cold" to "hot" and stimulating a systemic anti-tumor immune response. The combination leverages the immunogenic cell death induced by both Olvi-Vec and platinum-based chemotherapy to enhance the efficacy of immune checkpoint inhibitors, while also potentially reversing resistance or refractoriness to platinum-based agents.
