Overview
Prilosec + Pepcid AC (also known as OX17) is an investigational fixed-dose combination (FDC) therapy developed by Orexo AB, combining the proton pump inhibitor (PPI) omeprazole and the H2 receptor antagonist (H2RA) famotidine. This dual-mechanism approach is designed to provide more rapid and comprehensive gastric acid suppression than either agent alone. Famotidine provides a fast onset of action by competitively blocking histamine H2 receptors on gastric parietal cells, while omeprazole provides sustained acid control by irreversibly inhibiting the H+/K+-ATPase proton pump. The combination was primarily evaluated in Phase 2 clinical trials for the treatment of gastroesophageal reflux disease (GERD) and frequent heartburn, with studies focusing on 24-hour intragastric pH profiles to demonstrate superior acid control compared to monotherapy.
