Omeprazole + ranitidine is a combination of two pharmaceutical agents used to reduce gastric acid secretion. Omeprazole is a proton pump inhibitor (PPI) that irreversibly inhibits the H+/K+ ATPase enzyme system (proton pump) in the gastric parietal cells, leading to profound and prolonged reduction of stomach acid production. Ranitidine is a histamine H2-receptor antagonist that blocks histamine action at the H2 receptors of the gastric parietal cells, thereby reducing both basal and stimulated gastric acid secretion. This combination has been studied for use in conditions such as gastroesophageal reflux disease (GERD) and peptic ulcer disease, particularly for patients who experience nocturnal acid breakthrough or are refractory to standard therapy. Clinical studies suggest that while this combination can provide enhanced suppression of gastric acidity compared to monotherapy with either agent alone, its clinical benefit over optimized PPI therapy remains uncertain[1][3][5]. Ranitidine has been withdrawn from many markets due to concerns about NDMA contamination[4].
