Overview

OnabotulinumtoxinA + baclofen is a combination therapy used primarily for the management of spasticity, particularly in conditions such as cerebral palsy and spinal cord injury. OnabotulinumtoxinA (commonly known by the brand name Botox) is a neurotoxic protein that inhibits acetylcholine release at neuromuscular junctions, resulting in localized muscle relaxation and reduced spasticity. Baclofen is a gamma-aminobutyric acid (GABA) agonist that acts centrally to inhibit excitatory neurotransmitter release, reducing muscle tone and spasms. The combination leverages the focal effect of onabotulinumtoxinA with the generalized antispastic action of baclofen—often administered intrathecally—to improve overall control of spasticity, pain, quality of life, and self-care in affected patients[1][3][7].
