A multi-drug combination regimen that comprises **onartuzumab** (a monovalent monoclonal antibody targeting the MET receptor), **bevacizumab** (a monoclonal antibody targeting vascular endothelial growth factor A/VEGF-A), **paclitaxel** (a microtubule-stabilizing cytotoxic small molecule), and a **platinum-based chemotherapy agent** (cisplatin or carboplatin). This experimental combination has been most studied in first-line treatment of advanced **non-squamous non-small cell lung cancer (NSCLC)** and **triple-negative breast cancer (TNBC)** within randomized, multicenter, phase II trials. Onartuzumab inhibits the MET receptor pathway, bevacizumab inhibits angiogenesis, paclitaxel blocks cell division, and platinum agents crosslink DNA to induce apoptosis. The regimen generally uses intravenous administration. No significant benefit over current standards of care was demonstrated in NSCLC or TNBC, and onartuzumab addition was associated with higher rates of edema and thromboembolic events[1][3][5][7].
