Overview

This drug is a **combination regimen** consisting of five agents: - **Onartuzumab**: a monovalent monoclonal antibody targeting the extracellular domain of the MET receptor (hepatocyte growth factor receptor), designed to inhibit MET-driven tumor growth and oncogenic signaling. - **Folinic acid** (leucovorin): a reduced form of folic acid used to enhance the binding and cytotoxicity of fluorouracil. - **Fluorouracil** (5-FU): an antimetabolite and pyrimidine analog that inhibits thymidylate synthase, leading to impairment in DNA synthesis and function. - **Oxaliplatin**: a platinum-based chemotherapeutic that forms DNA crosslinks, inhibiting DNA replication and transcription. - **Bevacizumab**: a humanized monoclonal antibody against vascular endothelial growth factor A (VEGF-A), inhibiting angiogenesis required for tumor growth by preventing VEGF-A from binding to its receptors on endothelial cells. This combination has been explored in advanced solid tumors, specifically metastatic colorectal cancer and non-squamous non-small cell lung cancer, typically as a first-line therapy to improve response and survival compared to older regimens. The combination leverages simultaneous inhibition of oncogenic signaling, impairment of DNA synthesis and function, direct cytotoxic effects, and anti-angiogenic strategies.
