Overview

**Onartuzumab + vemurafenib** is a combination of two anticancer drugs: onartuzumab, a monoclonal antibody targeting the hepatocyte growth factor receptor (HGFR/MET), and vemurafenib, a small molecule inhibitor of the BRAF V600E mutant kinase. Onartuzumab acts by blocking MET signaling, thereby inhibiting tumor growth and metastasis in tumors where MET is upregulated. Vemurafenib selectively inhibits the activity of the BRAF V600E mutant kinase, which plays a critical role in the MEK/ERK signaling pathway, driving proliferation in certain cancers, particularly melanoma. This combination targets two complementary oncogenic pathways and would be evaluated primarily for cancers with both BRAF mutations and evidence of MET pathway activation.
