Overview

ONC-392 + pembrolizumab is an investigational combination therapy for cancer that pairs two immune checkpoint inhibitors. ONC-392 is a next-generation, humanized IgG1 monoclonal antibody targeting CTLA-4, designed to preserve CTLA-4 recycling and reduce toxicity while enhancing anti-tumor immune responses. Pembrolizumab (Keytruda) is a well-established monoclonal antibody targeting PD-1, blocking its interaction with PD-L1/PD-L2 to promote T-cell mediated anti-tumor activity. The combination aims to synergistically enhance immune activation against tumors by dual blockade of the CTLA-4 and PD-1 pathways. This regimen is being studied in advanced solid tumors—including non-small cell lung cancer and platinum-resistant ovarian cancer—where it seeks to improve efficacy over monotherapy or standard treatments[1][2][3][5][6].
