ONCOlogue (KRAS G12D) is a gene-silencing therapeutic candidate developed by Oncogenuity, designed to target DNA-level mutations in the KRAS gene. The drug utilizes the proprietary ONCOlogues platform, which employs peptide nucleic acids (PNAs) conjugated to cell-penetrating peptides (CPPs). Unlike traditional antisense or siRNA approaches that target mRNA, ONCOlogues are engineered to hybridize directly with mutant KRAS DNA sequences at the genomic level, suppressing transcription and selectively silencing mutant alleles such as G12D, G12C, and G12V while sparing wild-type KRAS. Currently in preclinical development, the program is being evaluated for the treatment of KRAS-driven malignancies, including pancreatic ductal adenocarcinoma, colorectal cancer, endometrial cancer, and non-small cell lung cancer (NSCLC). Preclinical studies have demonstrated selective repression of KRAS G12D mRNA and protein expression within hours of administration.
