Overview
ongericimab + toripalimab + bevacizumab + FOLFIRI is a combination therapy regimen being investigated for the treatment of advanced colorectal cancer, specifically in patients with microsatellite stable (MSS) or proficient mismatch repair (pMMR) status. This regimen is part of the ONTOP Phase II clinical trial sponsored by Shanghai Pudong Hospital. It combines ongericimab (a PCSK9 monoclonal antibody), toripalimab (a PD-1 immune checkpoint inhibitor), bevacizumab (an anti-angiogenic VEGF inhibitor), and the FOLFIRI chemotherapy backbone (irinotecan, leucovorin, and 5-fluorouracil). The combination aims to convert immunologically cold MSS colorectal tumors into hot tumors by leveraging the synergistic effects of immune checkpoint blockade, metabolic modulation via PCSK9 inhibition, and anti-angiogenesis, alongside traditional cytotoxic chemotherapy.
