Overview

This is a combination therapy consisting of ONO-7475, nivolumab, gemcitabine, and nab-paclitaxel. **ONO-7475** is a selective small molecule inhibitor of AXL and MER tyrosine kinases involved in survival and proliferation pathways in acute myeloid leukemia and potentially other malignancies, acting by inducing apoptosis through inhibition of several cell cycle regulators and anti-apoptotic proteins[1][2][4]. **Nivolumab** is a monoclonal antibody that inhibits the programmed death-1 (PD-1) immune checkpoint to enhance anti-tumor immune response. **Gemcitabine** is a nucleoside analog used as a cytotoxic chemotherapeutic agent, interfering with DNA synthesis. **Nab-paclitaxel** is an albumin-bound formulation of paclitaxel, which stabilizes microtubules and inhibits cell division, commonly used in solid tumors, especially pancreatic and breast cancers. The combination leverages multiple mechanisms—for example, ONO-7475’s kinase inhibition, nivolumab’s immunomodulation, and the antineoplastic/cytotoxic actions of gemcitabine and nab-paclitaxel—potentially resulting in additive or synergistic anti-tumor activity.
