Overview

OPC-167832 is a novel carbostyril derivative developed as an anti-tuberculosis agent. It acts by inhibiting decaprenylphosphoryl-β-D-ribose 2'-oxidase (DprE1), an essential enzyme for the biosynthesis of the mycobacterial cell wall, specifically arabinogalactan, which is crucial for Mycobacterium tuberculosis survival. Bedaquiline is a diarylquinoline antimycobacterial drug that inhibits mycobacterial ATP synthase, disrupting energy production in M. tuberculosis. The combination of OPC-167832 and bedaquiline has shown potent bactericidal activity in preclinical models and is being investigated in clinical trials for its potential to shorten therapy duration and improve outcomes in both drug-susceptible and multidrug-resistant tuberculosis[1][2][3][4][5].
